**Definition:** An antibody-drug conjugate (ADC) composed of a humanized monoclonal antibody directed against Ephrin-A4 (EFNA4), conjugated, via a hydrazone-based cleavable linker, to the DNA damaging agent calicheamicin, with potential antineoplastic activity. Upon administration of anti-EFNA4/calicheamicin ADC PF-06647263, the anti-EFNA4 monoclonal antibody moiety targets and binds to EFNA4 expressed on tumor cells. Upon binding, internalization and linker cleavage, calicheamicin is released and binds to the minor groove of DNA, causing double strand DNA breaks and may result in the inhibition of DNA synthesis and cell death in EFNA4-expressing tumor cells. EFNA4 is overexpressed by a variety of different cancer cell types and its expression is correlated with increased proliferation, invasion, and metastasis.
