**Definition:** A human monoclonal antibody directed against ST2, a receptor for the human inflammatory cytokine interleukin-33 (IL-33), with potential anti-inflammatory activity. Upon administration, astegolimab targets and binds to ST2. This prevents the binding of IL-33 to its receptor complex ST2 (IL1RL1) and IL-1 receptor accessory protein (IL-1RAcP). This inhibits the activation of IL-33/ST2-mediated signal transduction pathways. This may inhibit IL-33/ST2-mediated inflammation. IL-33 belongs to the IL-1 cytokine family and plays a key role in the modulation of inflammatory and neurodegenerative conditions. IL-33 is constitutively expressed at high levels in the nuclei of many cell types including endothelial, glial and some immune cells. It is involved in innate and adaptive immune responses via interaction with its receptor ST2, which is expressed on various immune cells, including macrophages, neutrophils, eosinophils, basophils, mast cells, type 2 helper T cells (Th2), regulatory T cells (Tregs), and group 2 innate lymphoid cells (ILC2s).
