**Definition:** A bifunctional monoclonal antibody targeting both the receptor tyrosine kinase epidermal growth factor receptor (EGFR) and the pro-inflammatory cytokine human transforming growth factor beta (TGF-beta; TGFb), with potential antineoplastic activity. Upon administration of ficerafusp alfa, the anti-EGFR moiety targets, binds to and prevents activation of EGFR-mediated signaling. This leads to an inhibition of EGFR-dependent downstream pathways and EGFR-dependent tumor cell proliferation and metastasis. The anti-TGFb moiety targets and binds to TGFb, thereby preventing the activation of TGFb-mediated signaling pathways. This may inhibit the proliferation of tumor cells in which TGFb is overactivated. EGFR and TGFb, mutated and/or overexpressed on the surfaces of various tumor cell types, play key roles in tumor cell proliferation and progression.
