Autologous CLL1-CAR-CD28-expressing T-lymphocytes

**Definition:** A preparation of autologous T-lymphocytes expressing a chimeric antigen receptor (CAR) specific for the tumor-associated antigen (TAA) C-type-lectin-like molecule-1 (CLL1; C-type lectin domain family 12 member A; CLEC12A) linked to the CD28 co-stimulatory signaling domain, with potential immunomodulating and antineoplastic activities. Upon administration, the autologous CLL1-CAR-CD28-expressing T-lymphocytes specifically target and bind to CLL1-expressing tumor cells. This induces selective toxicity in tumor cells that express the CLL1 antigen. CLL1, a member of the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily, is overexpressed in leukemic stem cells (LSCs) and plays an important role in disease progression and relapse for myeloid malignancies.
