**Definition:** An agonistic antibody targeting the human cell surface glycoprotein CD226 (DNAX accessory molecule-1; DNAM-1), with potential immunostimulatory and antineoplastic activities. Upon administration, anti-CD226 agonist antibody LY3435151 targets and binds to CD226 on a variety of immune cell types, including T-cells, natural killer (NK) cells, B-cells and monocytes. This induces CD226-dependent signaling pathways, which may trigger the activation of antigen-presenting cells (APCs) and activate T-cells and NK cells. This may result in an enhanced cytotoxic T-lymphocyte (CTL)-mediated and NK cell-mediated immune response against tumor cells. CD226, a cell surface glycoprotein that functions as an adhesion molecule, is expressed on various immune cells and plays an important role in the co-activation of T-cells and NK cells upon interaction with its ligands CD155 and CD112.
