Pegozafermin

**Definition:** A glycopegylated form of fibroblast growth factor 21 (FGF21), with metabolic regulating, anti-diabetic, anti-lipidemic, and anti-fibrotic activities. Upon administration, pegozafermin mimics FGF21, and binds to FGF21-specific receptors, FGFR1c, FGFR2c, and FGFR3c, in the liver and adipose tissue. This improves fat metabolism, reduces liver fat, improves insulin sensitization and insulin resistance, improves glycemic control, and protects hepatocytes from lipotoxicity-related stress. This may improve liver inflammation and fibrosis, resolve metabolic dysfunction-associated steatohepatitis (MASH), and decrease body weight and metabolic dysfunction. FGF21, primarily produced and secreted by hepatocytes in response to metabolic stressors, is an endocrine regulator of glucose and lipid metabolism and energy homoeostasis. Glycopegylation and site-specific mutations improve the half-life of pegozafermin over native FGF21.
