Acetaminophen/Tramadol

**Definition:** A combination preparation of the non-opiate, non-salicylate analgesic acetaminophen and the centrally acting synthetic opioid analgesic tramadol, with analgesic activity. Upon oral administration, acetaminophen exerts its analgesic activity by inhibiting prostaglandin synthesis, while tramadol exerts its analgesic activity by multiple mechanisms including the binding to the mu-opioid receptors in the central nervous system (CNS) and the weak inhibition of norepinephrine and serotonin reuptake.
