**Definition:** A preparation of off-the-shelf (OTS), allogeneic natural killer (NK) cells, derived from genetically engineered human induced pluripotent stem cells (iPSCs), that are genetically engineered to express a chimeric antigen receptor (CAR) specific for the tumor-associated antigen (TAA) cluster of differentiation 33 (CD33), and containing a high-affinity, non-cleavable CD16 (hnCD16) to enhance antibody-dependent cell-mediated cytotoxicity (ADCC), an interleukin-15 (IL-15) molecule to increase the persistence of allogeneic cells in vivo, and a CD38 knockout to avoid depletion of CAR-NK cells, with potential immunomodulating and antineoplastic activities. Upon administration, the allogeneic anti-CD33 CAR-NK cells QN-023a recognize, bind to and induce selective cytotoxicity in CD33-expressing tumor cells. CD33 is expressed on normal non-pluripotent hematopoietic stem cells and on myeloid leukemia cells.
