Anti-HLA-G Monoclonal Antibody UCB4594

**Semantic type:** Amino Acid, Peptide, or Protein|Immunologic Factor|Pharmacologic Substance

**Definition:** An afucosylated humanized immunoglobulin G1 (IgG1) monoclonal antibody targeting HLA-G histocompatibility antigen, class I, G (human leukocyte antigen G; HLA-G), with potential immune checkpoint inhibiting, immunomodulating and antineoplastic activities. Upon administration, the anti-HLA-G monoclonal antibody UCB4594 targets and binds to HLA-G, thereby preventing the binding of HLA-G to its inhibitory receptors immunoglobin-like transcript (ILT) 2 and ILT4 on a variety of immune cells, such as natural killer cells (NKs), T- and B-lymphocytes, and dendritic cells (DCs). This may prevent HLA-G-mediated immune suppression, thereby activating both innate and adaptive immune responses. This may activate anti-tumor immune responses. In addition, UCB4594 induces direct NK cell-mediated killing of tumor cells, complement dependent lysis and antibody-dependent cellular phagocytosis (ADCP). HLA-G, an immune checkpoint normally expressed at the maternal-fetal interface, is expressed across multiple tumor types and plays a key role in cancer immune evasion.

**Synonyms:** - UCB 4594 - UCB-4594 - UCB4594
