**Definition:** A humanized immunoglobulin G4 (IgG4) monoclonal antibody directed against interleukin-18-binding protein (IL18BP; IL-18BP), with potential immunostimulatory and antineoplastic activities. Upon administration, anti-IL-18BP monoclonal antibody GS-0321 targets and binds to IL-18BP. This results in the release of the pro-inflammatory cytokine IL-18 from pre-formed IL-18:IL-18BP complex. IL-18 binds to and activates the IL-18 receptor, which activates T-cells and natural killer (NK) cells and induces the secretion of interferon-gamma (IFNg). This potentiates the immune response against tumor cells. IL-18BP acts as a decoy receptor of IL-18 and prevents the activation of IL-18 receptor in the tumor microenvironment (TME).
