FGFR3/4 Inhibitor TYRA-430

**Definition:** An orally bioavailable reversible inhibitor of fibroblast growth factor receptor (FGFR) types 3 and 4 (FGFR3/4), with potential antineoplastic activity. Upon oral administration, FGFR3/4 inhibitor TYRA-430 targets, reversibly binds to and inhibits FGFR3/4. This blocks the signaling through these receptors mediated by the ligand fibroblast growth factor 19 (FGF19), thereby inhibiting tumor cell proliferation in FGF19- and FGFR4-overexpressing tumor cells. FGFR, a family of receptor tyrosine kinases upregulated in many tumor cell types, plays a key role in cellular proliferation, cell survival and angiogenesis. FGF19 is overexpressed by certain tumor cell types.
