Anti-ROR1 Antibody-drug Conjugate IM-1021

**Semantic type:** Pharmacologic Substance

**Definition:** An antibody-drug conjugate (ADC) composed of an immunoglobulin G1 (IgG1) monoclonal antibody directed against the tumor-associated antigen (TAA) receptor tyrosine kinase-like orphan receptor 1 (ROR1) conjugated, via a cleavable linker, to the camptothecin derivative and topoisomerase 1 inhibitor HC74, with potential antineoplastic activity. Upon administration, the monoclonal antibody moiety of anti-ROR1 ADC IM-1021 targets and binds to ROR1 expressed on tumor cells. Upon binding, internalization and binder cleavage, HC74 is released. HC74 inhibits DNA topoisomerase I activity, thereby inhibiting DNA replication and resulting in cell cycle arrest and tumor cell apoptosis. This inhibits the proliferation of ROR1-expressing tumor cells. IM-1021 is also able to induce a bystander effect on neighboring cells in the tumor environment. This further inhibits the proliferation of tumor cells. ROR1, also known as neurotrophic tyrosine kinase, receptor-related 1 (NTRKR1), is expressed during embryogenesis and in various hematological and solid malignancies. It plays key roles in tumor cell proliferation and survival.
