Anti-CLDN1 Antibody-drug Conjugate ALE.P02

**Definition:** An antibody-drug conjugate (ADC) composed of a monoclonal antibody directed against a specific epitope of the tumor-associated antigen (TAA) claudin1 (CLDN1; CLDN-1; claudin-1), conjugated, via a linker, to a tubulin inhibitor payload, with potential antineoplastic activity. Upon administration, anti-CLDN1 ADC ALE.P02 specifically targets and binds to the CLDN1 epitope expressed on tumor cells. Upon internalization, the tubulin inhibitor targets and binds to tubulin, and inhibits microtubule polymerization. This results in G2/M phase cell cycle arrest and apoptosis in CLDN1-expressing tumor cells. CLDN1 is overexpressed on a variety of tumor cells where it is exposed outside of the normal junctions. Its expression in healthy cells is hidden in tight junctions.
