Anti-TM4SF1 Antibody-drug Conjugate AGX101

**Definition:** An antibody-drug conjugate (ADC) composed of a monoclonal antibody directed against the tumor-associated antigen (TAA) transmembrane 4 L six family member 1 (TM4SF1) site-specifically conjugated to a maytansinoid tubulin inhibitor, with potential antineoplastic activity. Upon administration of anti-TM4SF1 ADC AGX101, the antibody moiety targets and binds to TM4SF1 expressed on tumor cells and tumor-associated vascular endothelium. Upon binding and the internalization of TM4SF1 via microtubules to the nucleus, the maytansinoid tubulin inhibitor is delivered to the nucleus of tumor cells. The maytansinoid binds to tubulin and disrupts microtubule assembly/disassembly dynamics, resulting in the inhibition of both cell division and cell growth of TM4SF1-expressing tumor cells and tumor-associated vascular endothelium. TM4SF1 plays a key role in the regulation of cell development, activation, growth and motility, and is overexpressed by a variety of tumor cells.
