Pan-KRAS Inhibitor JAB-23E73

**Definition:** An orally bioavailable inhibitor of both the active, guanosine triphosphate (GTP)-bound (ON) form and the inactive, guanosine diphosphate (GDP)-bound (OFF) form of various oncogenic KRAS mutations, with potential antineoplastic activity. Upon oral administration, pan-KRAS inhibitor JAB-23E73 targets, binds to and inhibits various forms of KRAS mutations, thereby inhibiting KRAS-dependent signaling. This inhibits the proliferation of and induces apoptosis in tumor cells in which KRAS is mutated. KRAS plays an important role in cell signaling, division and differentiation. Mutations of KRAS may induce constitutive signal transduction leading to tumor cell growth, proliferation, invasion, and metastasis.
