CCR2-targeting Chimera STX-0712

**Definition:** A C-C chemokine receptor type 2 (CCR2)-targeting chimera, composed of a bifunctional small molecule with a CCR2-binding moiety and a linker that conjugates to an antibody-dependent cellular cytotoxicity (ADCC)-enhanced antibody, with potential antineoplastic activity. Upon administration of CCR2-targeting chimera STX-0712, the CCR2-binding moiety targets and binds to CCR2 expressed on malignant monocytes, and the antibody moiety induces ADCC against the CCR2-expressing malignant monocytes, thereby killing malignant monocytes. The G-protein coupled receptor CCR2 is expressed on the surface of monocytes and macrophages, and stimulates their migration and infiltration; it plays a key role in inflammation. CCR2 is overexpressed in certain cancers, including chronic myelomonocytic leukemia (CMML), and it is involved in angiogenesis, tumor cell migration and proliferation.
