Engineered Cytokine ORB-021

**Definition:** An engineered cytokine, with potential immunomodulating and antineoplastic activities. Upon administration, the engineered cytokine ORB-021 is inactive and, upon binding to PD-L1-expressing tumor and immune cells, becomes activated. This results in the activation of natural killer (NK) and dendritic cells (DCs), and inhibits myeloid derived suppressor cells (MDSCs) in the tumor microenvironment (TME). This may result in an anti-tumor immune response and tumor cell killing. ORB-021 is inactive and becomes activated only when bound to its target cells, thereby enabling in-cis on-target signaling. This minimizes off-target interactions as seen with traditional cytokine therapy.
