Vemzatatug Vedotin

**Definition:** An antibody-drug conjugate (ADC) composed of vemzatatug, a monoclonal antibody directed against the human G-protein coupled receptor family C group 5 member D (GPRC5D) conjugated to the auristatin derivative and microtubule-disrupting cytotoxic agent monomethyl auristatin E (MMAE), with potential antineoplastic activity. Upon administration of vemzatatug vedotin, the anti-GPRC5D antibody moiety selectively binds to tumor cells expressing GPRC5D. Upon binding and internalization, MMAE binds to tubulin and inhibits its polymerization, which results in G2/M phase arrest and induces apoptosis in GPRC5D-expressing tumor cells. In addition, AZD0305 induces an antibody-dependent cell-mediated cytotoxicity (ADCC) against GPRC5D-expressing tumor cells, thereby further killing the tumor cells. GPRC5D is overexpressed on certain tumors, such as multiple myeloma, while minimally expressed on normal, healthy cells, and plays a key role in tumor cell proliferation.
