Anti-CD37/DM1 Derivative Antibody-drug Conjugate Debio 1562M

**Definition:** An antibody-drug conjugate (ADC) composed of naratuximab, a humanized immunoglobulin G1 (IgG1) monoclonal antibody directed against the tumor-associated antigen (TAA) CD37, covalently linked, via a protease cleavable linker, to a derivative of the maytansinoid mertansine (DM1), a microtubule disrupting agent, with potential antineoplastic activity. Upon administration of anti-CD37/DM1 derivative ADC Debio 1562M, the naratuximab moiety targets and binds to CD37 on tumor B-cells. Upon internalization and linker cleavage by cathepsin B, the DM1 derivative is released, binds to tubulin and inhibits tubulin polymerization and microtubule assembly, resulting in G2/M checkpoint arrest and apoptosis in CD37-expressing tumor cells. CD37, a transmembrane glycoprotein, is overexpressed in B-cell malignancies.
