Anti-mesothelin T-cell Engaging Bispecific Antibody CT-95

**Definition:** A bispecific T-cell engager (TCE) and humanized Fc-silenced bispecific antibody with two moderate affinity mesothelin (MSLN)-binding arms directed against a membrane-proximal epitope of the tumor-associated antigen (TAA) MSLN and two CD3-binding single chain variable fragment (scFv) domains, with potential immunostimulating and antineoplastic activities. Upon administration, anti-mesothelin T-cell engaging bispecific antibody CT-95 targets and binds to MSLN expressed on tumor cells and to CD3 expressed on cytotoxic T-lymphocytes (CTLs). This activates and redirects CTLs to MSLN-expressing tumor cells, which results in the CTL-mediated death of MSLN-expressing tumor cells. MSLN, a cell surface glycoprotein involved in cell adhesion, is overexpressed in a variety of cancer cell types. By targeting the membrane-proximal epitope of MSLN, CT-95 may be able to avoid unwanted binding to shed MSLN peptides in the tumor environment (TME).
