Anti-CDH17 Antibody-drug Conjugate YL217

**Definition:** An antibody-drug conjugate (ADC) composed of a humanized immunoglobulin G1 (IgG1) monoclonal antibody targeting the tumor-associated antigen (TAA) cadherin-17 (CDH17) conjugated, via a cleavable methylsulfonyl pyrimidine tripeptide linker, to the topoisomerase-1 inhibitor (TOP1i) YL0010014, with potential antineoplastic activity. Upon administration of anti-CDH17 ADC YL217, the antibody moiety targets and binds to CDH17-expressing tumor cells. Upon binding, internalization and linker cleavage, the topoisomerase-1 inhibitor YL0010014 is released and inhibits DNA topoisomerase I activity, thereby inhibiting DNA replication and resulting in cell cycle arrest and apoptosis in tumor cells expressing CDH17. CDH17, a cell surface adhesion protein, is overexpressed in a variety of tumors of the gastrointestinal (GI) tract.
