KRAS G12D Inhibitor VS-7375

**Definition:** An orally bioavailable non-covalent small molecule inhibitor of both the active, guanosine triphosphate (GTP)-bound form and the inactive, guanosine diphosphate (GDP)-bound form of the oncogenic KRAS substitution mutation G12D, with potential antineoplastic activity. Upon oral administration, KRAS G12D inhibitor VS-7375 specifically targets, binds to, and inhibits both the GTP-bound (active) and GDP-bound (inactive) forms of KRAS G12D. This prevents KRAS G12D(ON)-mediated signaling and the activation of downstream survival pathways, which leads to an inhibition of the growth of tumor cells that overexpress KRAS G12D. KRAS, a member of the RAS family of oncogenes, serves an important role in cell signaling, division and differentiation. Mutations of KRAS may induce constitutive signal transduction leading to tumor cell proliferation, invasion, and metastasis.
