Anti-CG1/Anti-CD3 Bispecific T-cell Engager CBX-250

**Definition:** A T-cell receptor-mimetic (TCRm) bispecific T-cell engager (TCE) antibody directed against the CG1/HLA-A*02:01 peptide-major histocompatibility complex (pMHC) complex and the T-cell surface antigen CD3, with potential immunostimulating and antineoplastic activities. Upon administration, anti-CG1/anti-CD3 bispecific TCE CBX-250 targets and binds to both the CG1/HLA-A*02:01 pMHC complex on tumor cells and CD3 on T-cells. This results in the cross-linking of tumor cells and T-cells, and induces a cytotoxic T-lymphocyte (CTL) response against CG1/pMHC-positive tumor cells. CBX-250 is also able to induce a bystander killing effect on neighboring CG1/pMHC-negative cells in the tumor environment. Cathepsin G (CG), an intracellular serine protease, is overexpressed and ubiquitinated in acute myeloid leukemia (AML) blasts and leukemic stem cells. CG1 (FLLPTGAEA) is an HLA-A*02:01-restricted cathepsin G peptide.
