Pan-RAS Molecular Glue ERAS-0015

**Definition:** An orally bioavailable molecular glue and inhibitor that targets the oncogenic RAS wild-type (WT) and various RAS mutant forms, including G12X/G13X/Q61X mutations, with potential antineoplastic activity. Upon oral administration, pan-RAS molecular glue ERAS-0015 selectively targets and binds to RAS in the guanosine triphosphate (GTP)-bound (active) state and cyclophilin A (CYPA), forming a ternary complex, thereby inhibiting cellular signaling mediated by both mutant and wild-type RAS. This inhibits the growth and survival of RAS-overexpressing and mutated tumor cells. RAS plays an important role in cell signaling, division, differentiation, and proliferation. Mutations of RAS may induce constitutive signal transduction leading to tumor cell growth, proliferation, invasion, and metastasis.
