**Definition:** An antibody-drug conjugate (ADC) composed of a humanized monoclonal antibody directed against the SEA domain of the tumor associated antigen (TAA) mucin-1 (MUC1) conjugated to an antimitotic cytotoxic payload via a linker, with potential antineoplastic activity. Upon administration, the antibody moiety of anti-MUC1-SEA ADC SBO-154 targets and binds to the SEA domain of MUC1 (MUC1-SEA) expressed on tumor cells. Upon binding, the antimitotic payload disrupts mitosis and inhibits cell proliferation in tumor cells expressing MUC1. MUC1, a glycoprotein overexpressed on the surface of a variety of cancer cells, plays a key role in tumor cell survival and proliferation. MUC1-SEA is expressed at very low levels in plasma, and targeting MUC1-SEA prevents the binding of the ADC to MUC1-variable number tandem repeat (VNTR) antigens shed in peripheral blood and thus allows the ADC to preferentially target MUC1 expressed on tumor cells.
