**Definition:** An antibody-drug conjugate (ADC) composed of a monoclonal antibody directed against integrin beta-6 (IB6) conjugated, via a cleavable glucuronide linker, to the camptothecin analog, 7-aminomethyl-10,11-methylenedioxycamptothecin (AMDCPT), with potential antineoplastic activity. Upon administration, the antibody moiety of anti-IB6 ADC PF-08046876 targets and binds to IB6 expressed on the surface of tumor cells. Upon binding and linker cleavage, AMDCPT is released, and inhibits DNA topoisomerase-1 activity, thereby inhibiting DNA replication and resulting in cell cycle arrest and tumor cell apoptosis. This inhibits the proliferation of IB6-expressing tumor cells. IB6 is overexpressed in certain cancer cell types and has been associated with increased proliferation, migration and invasion of tumor cells.
