Anti-CD22/Topoisomerase I Inhibitor Antibody-drug Conjugate AZD4512

**Definition:** An antibody-drug conjugate (ADC) composed of a monoclonal antibody directed against the tumor-associated antigen (TAA) CD22 conjugated, via cleavable linker, to a DNA topoisomerase I inhibitor (TOP1i), with potential antineoplastic activity. Upon administration, anti-CD22/TOP1i ADC AZD4512 targets and binds to CD22 expressed on tumor cells. Upon binding, internalization, and linker cleavage, the TOP1i is released and inhibits DNA topoisomerase I activity, thereby inhibiting DNA replication and resulting in DNA damage, cell cycle arrest and apoptosis in tumor cells overexpressing CD22-expressing tumor cells. CD22, a B-lineage-restricted, transmembrane phosphoglycoprotein, is expressed on mature B-cells and on most malignant B-cells.
