Anti-nectin-4/Anti-TROP2 Bispecific Antibody-drug Conjugate AVZO-103

**Definition:** An antibody-drug conjugate (ADC) composed of a bispecific antibody directed against the tumor associated antigens (TAAs) nectin-4 (PVRL4) and trophoblast cell surface protein 2 (trophoblast antigen 2; calcium signal transducer 2; TROP2; TROP-2; TACSTD2; GA733-1; M1S1) conjugated to an as of yet undisclosed cytotoxic payload, with potential antineoplastic activity. Upon administration, the antibody moiety of anti-nectin-4/anti-TROP2 bispecific ADC AVZO-103 targets and binds to nectin-4 and TROP2 expressed on tumor cells. Upon binding and the release of the cytotoxic payload, nectin-4/TROP2-expressing tumor cells are killed through an as of yet unelucidated mechanism of action. Nectin-4 and TROP2 are co-expressed in a variety of cancer cells. Nectin-4, a TAA belonging to the nectin family, is overexpressed in a variety of cancers, but has a restricted distribution in normal tissue. TROP2, a transmembrane protein overexpressed in various tumors, is associated with enhanced tumor aggressiveness, metastasis, drug resistance and increased tumor cell survival.
