Anti-CCR8 Antibody-drug Conjugate TAK-188

**Definition:** An antibody-drug conjugate (ADC) composed of an antibody directed against human C-C motif chemokine receptor 8 (CCR8) conjugated to an as of yet undisclosed cytotoxic payload, with potential immunomodulating and antineoplastic activities. Upon administration, the antibody moiety of anti-CCR8 ADC TAK-188 targets and binds to CCR8 on CCR8-positive, immunosuppressive tumor-infiltrating T regulatory (TITR) cells in the tumor microenvironment (TME). Upon binding, CCR8-expressing TITR cells are killed through an as of yet unelucidated mechanism of action. This depletes CCR8-positive TITR cells, which may reverse the suppression of CD8+ effector T-cells and reactivate antitumor immune responses. CCR8 is specifically expressed by TITR cells in multiple types of cancer and plays a key role in immunosuppression.
