Anti-claudin-1/TOP1i Antibody-drug Conjugate ALE.P03

**Definition:** An antibody-drug conjugate (ADC) composed of a monoclonal antibody directed against the tumor-associated antigen (TAA) claudin1 (CLDN1; CLDN-1; claudin-1) conjugated, via a linker, to a DNA topoisomerase I inhibitor (TOP1i), with potential antineoplastic activity. Upon administration, anti-CLDN1/TOP1i ADC ALE.P03 targets and binds to CLDN1 expressed on tumor cells. Upon internalization, the TOP1i inhibits DNA topoisomerase I activity, thereby inhibiting DNA replication and resulting in cell cycle arrest and apoptosis in CLDN1-expressing tumor cells. CLDN1 is overexpressed on a variety of tumor cells where it is exposed outside of the normal junctions. Its expression in healthy cells is hidden in tight junctions.
