**Definition:** An antibody-drug conjugate (ADC) composed of Mab0727, a monoclonal antibody targeting the tumor-associated antigen (TAA) cadherin-17 (CDH17) conjugated, via a cleavable linker, to the topoisomerase-1 inhibitor (TOP1i) MF-6, with potential antineoplastic activity. Upon administration of anti-CDH17/MF-6 ADC 7MW4911, Mab0727 selectively targets and binds to CDH17-expressing tumor cells. Upon binding, internalization and linker cleavage, MF-6 is released and inhibits DNA topoisomerase I activity, thereby inhibiting DNA replication and resulting in cell cycle arrest and apoptosis in tumor cells expressing CDH17. CDH17, a member of the 7D-cadherin superfamily and a cell surface adhesion protein, is overexpressed in a variety of tumors of the gastrointestinal (GI) tract.
