Anti-CDH17/Exatecan Antibody-drug Conjugate ARR-217

**Definition:** An antibody-drug conjugate (ADC) composed of a humanized immunoglobulin G1 (IgG1) monoclonal antibody targeting the tumor-associated antigen (TAA) cadherin-17 (CDH17) site-specifically conjugated to the topoisomerase-1 inhibitor exatecan, with potential antineoplastic activity. Upon administration, anti-CDH17/exatecan ADC ARR-217 selectively targets and binds to CDH17-expressing tumor cells. Upon binding and internalization, exatecan is released and inhibits DNA topoisomerase 1 activity, thereby inhibiting DNA replication and resulting in cell cycle arrest and apoptosis in tumor cells expressing CDH17. CDH17, a member of the 7D-cadherin superfamily and a cell surface adhesion protein, is overexpressed in a variety of tumors of the gastrointestinal (GI) tract.
