CD8-guided IL-2 Fusion Protein AZD6750

**Definition:** A recombinant fusion protein comprised of two CD8 binding domains and two IL-2 mutein domains, with potential immunostimulating and antineoplastic activities. Upon administration, CD8-guided IL-2 fusion protein AZD6750 specifically targets and preferentially binds, with its CD8 binding domains, to CD8alpha on CD8+ T-cells, thereby allowing for the IL-2 mutein domains to specifically bind to the CD122/CD132 IL-2R complex, which activates IL-2R-mediated signaling in CD8+ T-cells. This enhances cytotoxic T-lymphocytes (CTLs)-mediated cytotoxic immune responses against tumor cells. The IL-2 mutein has a reduced affinity to IL-2 receptor subunit alpha (IL-2Ralpha; CD25), which promotes the selective activation of CD8+ T-cells over the immunosuppressive regulatory T-cells (Tregs). The CD8-targeted delivery of IL-2 allows for reduced systemic toxicity while enhancing CD8-mediated tumor cell killing.
