Anti-Claudin 6 Antibody-drug Conjugate AT03-65

**Definition:** An antibody-drug conjugate (ADC) consisting of a recombinant monoclonal antibody directed against the tumor-associated antigen (TAA) claudin 6 (CLDN6; CLDN-6) conjugated, via a cleavable and hydrophilic linker, to a derivative of the cytotoxic DNA alkylating agent trabectedin, with potential antineoplastic activity. Upon administration of anti-CLDN6 ADC AT03-65, the anti-CLDN6 antibody moiety targets and binds to CLDN6-expressing tumor cells. Upon binding, internalization and linker cleavage, the trabectedin derivative is released, and binds to and alkylates DNA, thereby causing cell cycle arrest, apoptosis and inhibition of cell growth in CLDN6-expressing tumor cells. CLDN6, a transmembrane tight-junction protein and embryonic antigen, is overexpressed on a variety of tumor cells but is not expressed on normal, healthy adult cells.
