Anti-nectin-4/CD28 Bispecific Antibody RNDO-564

**Definition:** An immunoglobulin G (IgG)-like human tumor-targeted co-stimulatory bispecific antibody directed against the tumor-associated antigen (TAA) nectin-4 (PVRL4) and the co-stimulatory T-cell surface glycoprotein CD28, with potential immunostimulating and antineoplastic activities. Upon administration, anti-nectin-4/CD28 bispecific antibody RNDO-564 targets and binds to both nectin-4 expressed on certain tumor cells and CD28 on activated T-lymphocytes. This allows for conditional stimulation and activation of CD28 and results in T-cell stimulation and enhancement of T-lymphocyte-mediated cytotoxicity and cytokine release, which may result in the CTL-mediated cell death of nectin-4-expressing tumor cells. In addition, RNDO-564 induces bystander killing of nectin-4-negative tumor cells. Nectin-4, a cell adhesion molecule belonging to the nectin family, is overexpressed on certain tumor cell types while it has a restricted distribution in normal tissue. CD28 co-stimulation is essential for sustained T-cell function and T-cell-mediated killing while preventing T-cell exhaustion within the immunosuppressive tumor microenvironment (TME). By delivering tumor-targeted CD28 agonism, RNDO-564 enhances anti-tumor T-cell responses while avoiding systemic toxicity.
