**Definition:** A recombinant, engineered human immunoglobulin G1 (IgG1) Fc fragment containing a F241A substitution, with potential immunomodulating and anti-inflammatory activities. The F241A substitution induces the same conformational change as sialylation of IgG antibodies. Upon intravenous administration, human IgG1 Fc-F241A fragment NVG-2089, mimicking sialylated IgG, targets and binds to immunomodulatory type II Fcgamma receptors. This binding upregulates the expression of the inhibitory immune checkpoint FcgRIIb on B-cells and myeloid cells and causes the expansion of T-regulatory cells, downregulates effector T-cells, and downregulates the expression of inflammatory genes involved in numerous inflammatory pathways. This reduces autoimmune dysregulation, reduces the dysregulated inflammatory response, and induces immune tolerance in various diseases, such as chronic inflammatory demyelinating polyneuropathy (CIDP) and immune thrombocytopenia (ITP). Non-sialylated IgGs bind, upon binding to and complex formation with antigens, to pro-inflammatory type I Fcgamma receptors, causing an inflammatory response.
