Anti-DLL3/Exatecan Antibody-drug Conjugate IBI3009

**Definition:** An antibody-drug conjugate (ADC) composed of a monoclonal antibody directed against the tumor-associated antigen (TAA) delta-like protein 3 (DLL3) conjugated, via a cleavable linker, to the camptothecin derivative and topoisomerase 1 inhibitor exatecan, with potential antineoplastic activity. Upon administration of anti-DLL3/exatecan ADC IBI3009, the monoclonal antibody moiety targets and binds to DLL3-expressing tumor cells. Upon binding and linker cleavage, exatecan is released and inhibits DNA topoisomerase I activity, thereby inhibiting DNA replication and resulting in cell cycle arrest and apoptosis in tumor cells expressing DLL3. DLL3, a Notch pathway protein, is overexpressed on a variety of cancer cell types. It plays a key role in embryonic development, tumor initiation and proliferation.
