Non-muscle Myosin IIA/IIB Inhibitor MT-125

**Definition:** A selective, central nervous system (CNS)-permeable inhibitor of both non-muscle myosin II (NMII) A and B, with potential antineoplastic activity. Upon administration, NMIIA/IIB inhibitor MT-125 targets and binds to both NMIIA and NMIIB, thereby blocking their activation and NMIIA/B-mediated signaling. This simultaneously inhibits tumor cell invasion and metastasis, and tumor cell proliferation. In addition, MT-125 generates reactive oxygen species (ROS) within tumor cells, thereby inducing redox stress and DNA damage in tumor cells. MT-125 also enhances the platelet-derived growth factor receptor (PDGFR)-mediated signaling pathway. NMIIA and NMIIB, motor proteins that act on actin to regulate cytoskeletal function, play crucial roles in movement, division, signaling and mitochondrial dynamics. Upregulated in certain tumor cells, NMIIA regulates cell movement by physically moving and reshaping the actin cytoskeleton, and allows cancer cells to change shape to promote the invasion of healthy tissue while NMIIB regulates cell division, and promotes tumor cell proliferation.
