**Semantic type:** Molecular Biology Research Technique
**Definition:** Replacement of a gene by an altered version of the same gene using homologous recombination resulting in a gain of function. One example of a knock-in is deletion of a coding sequence of a gene in a mouse and then replacing it with human coding sequences.
