LVV Expressing VSV Glycoprotein/Anti-CD3 scFv Containing Transgene Encoding for Anti-BCMA CAR KLN-1010

**Definition:** A gene therapy agent composed of a replication-incompetent, self-inactivating lentiviral vector (LVV) expressing a modified vesicular stomatitis virus (VSV) glycoprotein fusogen and an anti-CD3 single chain variable fragment (scFv), and encapsulating a transgene encoding for a chimeric antigen receptor (CAR) that targets the tumor-associated antigen (TAA) B-cell maturation antigen (BCMA; tumor necrosis factor receptor superfamily member 17; TNFRSF17), with potential to generate anti-BCMA CAR T-cells in vivo that may have immunostimulating and antineoplastic activities. Upon administration of LVV expressing VSV glycoprotein/anti-CD3 scFv containing transgene encoding for anti-BCMA CAR KLN-1010, the anti-CD3 scFv and VSV glycoprotein mediate CD3-positive T-cell binding, activation and T-cell transduction. Upon transduction, the T-cells express an anti-BCMA CAR. These in vivo generated anti-BCMA CAR T-cells induce cytokine production, proliferate and kill BCMA-expressing tumor cells. BCMA, a receptor for both a proliferation-inducing ligand (APRIL) and B-cell activating factor (BAFF), is a member of the tumor necrosis factor receptor superfamily (TNFRSF). BCMA is found on the surfaces of plasma cells, is overexpressed on malignant plasma cells and plays a key role in plasma cell proliferation and survival.
