Adeno-associated Viral Vector Encoding Anti-HER2/CD3 Bispecific T-Cell Engager VNX-202

**Definition:** An adeno-associated virus (AAV) encoding GP202, a single-chain variable fragment (scFv) diabody and bispecific T-cell engager (TCE) targeting the tumor-associated antigen (TAA) human epidermal growth factor receptor 2 (EGFR2; HER2; ErbB2) and cluster of differentiation (CD) 3 (CD3) on T-cells, with potential immunostimulating and antineoplastic activities. Upon intravenous administration, AAV encoding anti-HER2/CD3 bispecific TCE VNX-202 induces the continuous secretion of GP202 by hepatocytes into the bloodstream. This results in long-term, consistent serum levels of GP202. GP202 binds to both the CD3 antigen on T-cells and the HER2 antigen expressed on tumor cells. This results in the cross-linking of T-cells and tumor cells, and induces a cytotoxic T-lymphocyte (CTL) response against HER2-expressing tumor cells. HER2 is overexpressed in a variety of cancer cell types and is associated with increased tumor cell proliferation.
