Anti-CD37/DXd Antibody-drug Conjugate DS3790

**Definition:** An antibody-drug conjugate (ADC) composed of a humanized immunoglobulin G1 (IgG1) monoclonal antibody directed against the tumor-associated antigen (TAA) CD37 conjugated, via a tetrapeptide-based cleavable linker, to the cytotoxic DNA topoisomerase I inhibitor and exatecan derivative DXd (MAAA-1181a; MAAA-1181), with potential antineoplastic activity. Upon administration anti-CD37/DXd ADC DS3790, the antibody moiety targets and binds to CD37 on tumor B-cells. Upon binding, internalization and linker cleavage, the DXd moiety is released, binds to DNA topoisomerase I, and stabilizes the cleavable complex between topoisomerase I and DNA. This results in DNA breaks, inhibition of DNA replication and apoptosis in CD37-expressing tumor cells. CD37, a transmembrane glycoprotein, is overexpressed in B-cell malignancies.
