**Definition:** A preparation of autologous T-lymphocytes that have been genetically modified to express a chimeric antigen receptor (CAR) targeting the membrane-bound form of the tumor-associated antigen (TAA) human mucin-16 (MUC16, cancer antigen 125; CA125; FLJ14303) linked to the co-stimulatory domain 4-1BB (CD137) coupled to the zeta chain of the TCR/CD3 complex (TCRzeta; CD247; CD3zeta), and expressing the pro-inflammatory cytokine interleukin 18 (IL-18), with potential immunostimulating and antineoplastic activities. Upon administration, autologous anti-MUC16 CAR-BBz-IL-18-expressing T-cells GB-5267 recognize and bind to MUC16-expressing tumor cells, thereby inducing selective toxicity in MUC16-expressing tumor cells. MUC16, a member of the mucin family of glycoproteins, is overexpressed on a variety of cancer cell types. IL-18 promotes T- activation, proliferation, and persistence within the immunosuppressive tumor microenvironment (TME) and potentiates the immune response against tumor cells. GB-5267 targets the membrane-bound form of MUC16, thereby selectively targeting MUC16-expressing tumor cells and avoiding shed MUC16 circulating in the blood.
