Single Protein Encapsulated Doxorubicin SPEDOX-6

**Definition:** A single protein encapsulation (SPE)-based drug formulation composed of nine molecules of the anthracycline antibiotic doxorubicin encapsulated within a single unmodified monomeric human serum albumin (HAS) molecule, with potential antineoplastic activity. Upon administration, SPE doxorubicin SPEDOX-6 is specifically taken up by fast proliferating tumor cells as HSA serves as a nutrient for tumor cells. Inside the tumor cell and upon release from HSA, doxorubicin intercalates into DNA and inhibits the topoisomerase II activity, which results in the inhibition of tumor cell DNA replication and tumor cell proliferation. Encapsulation of doxorubicin within HSA improves the pharmacokinetic (PK) profile of doxorubicin compared to unformulated doxorubicin with improved drug penetration specifically into tumors over healthy cells due to both the high permeability and retention effects of the tumor vasculature. This improves efficacy and lowers the toxicity profile, including doxorubicin-induced cardiotoxicity.
