**Definition:** A nanoparticle-based formulation composed of the poorly water-soluble SN-38 (7-ethyl-10-hydroxy-camptothecin), a biologically active metabolite of the prodrug irinotecan, encapsulated within branched polymers composed of polyethyloxazoline (PEOX), with potential antineoplastic activity. Upon administration of the PEOX-based polymer encapsulated SN-38 FID-022, the nanoparticles accumulate at the tumor site, due to the unique characteristics of the tumor vasculature, while avoiding normal, healthy tissue. Once SN-38 is released, it binds to and inhibits topoisomerase I by stabilizing the cleavable complex between topoisomerase I and DNA, resulting in DNA breaks, inhibition of DNA replication, and apoptosis. Compared to the administration of SN-38 alone, this formulation increases SN-38's solubility and enhances its tumor tissue penetration and efficacy, while avoiding systemic exposure, which minimizes its toxicity.
